In Salesforce Marketing Cloud, all dates are stores in UTC format.

The date and time shown in DESelect Segment Filters is the date/time in UTC DESelect actually Filters on, no matter the timezone settings of the user using DESelect Segment.

For example, if you have an Order Date 'Thursday, December 05, 2019 12:00 AM' in your Data Extension, if you use the date picker in DESelect Segment on a Filter and you choose date the 5th of December 2019, and time 12:00AM, this record will be returned. This will be the same whether the user making this filter in DESelect Segment has his SFMC User Timezone setting set to New York or Amsterdam.

When a user changes the criteria of a selection, will DESelect modify the underlying Data Extension or will it create a new Data Extension?
